LEVEL SOME CYTOKINES IN BIOLOGICAL LIQUIDS AT CHILDREN, WHO HAVE BRONCHIAL ASTHMA

For last years growth bronchial asthma (BA) differs among the children in ecologically unpleasant region. Though the various aspects of a problem bronchial asthma are successfully developed, nevertheless, there are a lot of questions which require their decisions. In this plan represents the large scientific and practical interest study some cytokines in biological liquids at the patients of children BA. Last years in connection with occurrence of the new scientific items of information in clinical allergology and immunology, performance about pathogenesis of allergic reactions in general essentially has changed. And TH 1 and TH 2 play an important role in the development of the mechanism of allergological inflammation.  TH 1 is responsible for the cell development, and TH 2 hormonal immunity.   The basic role in the regulation immune answer between clones TH 1 and TH 2 crates play cytokines. The Rotting factor of a tumour α (TNFα), γ-interferon (INF-γ), interleukin 1β (IL-1β) and interleukin - 4 (IL-4).

MATERIALS AND METHODS
Definition of a level cytokines.  1 β (IL - 1β) and 4 (IL - 4), interferon γ (IFN γ), rotting factor of tumour (TNFα) determined by the method of immunoferment analysis.   St. Petersburg Scientific Research centers reactives are used.  48 patients who have bronchial asthma were surveyed.   27 of them were boys, 21 of them were girls.  The patients were divided into groups by the levels of occurrence. 
And 12 patients were in a status of a heavy attack, 14 attacks of average weight, 10 easy currents and 12 children in the period of remission of disease have made control group.  The figures have processed by a method of variational statistics.  Distinction are authentic at t ≥ 2, and p < 0.05.

RESULTS AND DISCUSSIONS
At 48 patients of children with bronchial asthma defined a level of the contents in whey of blood TNFα, IFN γ and IL - 1β.  12 patients were in a status of a heavy attack and 12 children, 14 attacks of average weight, of 10 easy attacks and 12 children in the period of remission of disease have made control group.  The received information demonstrates authentic dependence of concentration of a level against inflammation of cytokines in the whey of blood from weight of an attack bronchial asthma in the children. The revealed changes authentically differed from values received in group of comparison.

    So if at a heavy attack BA the level whey TNFα in the surveyed children was considerably exceeded (243.5±23.9 p2/ml) in comparison with the information in the children, who are in the period of remission – 82.4±7.0 p2/ml, p<0.001.   That at an attack of average weight is marked only moderate increase this cytokine (113.7±9.3 p2/ml) in comparison with remission p<0.05).  At an easy attack BA the level of TNFα, was comparable with whey concentration of cytokine in the period of remission that has made on the average – 89.3±6.3 p2/ml.  Analysis of results of research of a level of IL-1β in the whey of blood has shown, that in the children at heavy attack ВА is observed almost ten-multiple increase of its level in comparison with results at remission disease 346.7±36.6 p2/ml – against 35.8±3.9 p2/ml (p < 0.001).  In the children with an attack of average weight ВА the increase of a level of IL-1β more than 3 times was observed in comparison with group of children in the stage of remission – 110.4±5.2 pg/ml (p<0.001).  In the comparative analysis of parameters of children with an easy attack ВА (48.6±5.2 pg/ml) and children in a stage of remission (35.8±3.9 pg/ml) authentic distinctions was not observed. As known, IFN γ is produced TH 1 cells and NК cells.  In our researches is marked lowered in comparison with the group of children in a stage of remission. And this reduction is observed at an attack ВА of any degree of weight: at a heavy degree 74.3± 4.9 pg/ml (p<0.001), at an average degree of weight 78.5±7.3 pg/ml (p<0.001), and at an easy degree 75.8±5.7 pg/ml (p<0.001).  The degree of IFN γ in the period of remission was 131.7±11.0 pg/ml.  We carried out study cytokine structure of a secret mouth cavity in the children with bronchial asthma depending on weight current.  Analyzed the contents three types of cytokine - IL-1β,  TNFα, and IL-4.

In the group of healthy children the contents of IL-1β in saliva has made 21.8±1.80 pg/ml.  In groups of the patients the following results are received:  heavy current 196.0±20.76 pg/ml, middle heave 128.0±14.04 pg/ml and easy current 99.7±10.09 pg/ml, (for all parameters p<0.001).  The distinctions between groups with various weight of current are authentic (p<0.001).  The contents TNFα in saliva in the patients of control group has made 27.3±2.55 pg/ml.  In other groups of the patients the following results are received: heavy current 95.7±9.16 pg/ml, middle heave 54.6±4.56 pg/ml and easy current 47.2±4.84 pg/ml, (for all parameters p<0.01-0.001).  In the patients of control group the contents of IL-4 in saliva has made 6.2±0.41 pg/ml.  In the group of the patients the following results were received: heavy current 19.5±1.79 pg/ml, middle heave 12.7±1.02 pg/ml and easy current 14.9±1.39 pg/ml.
Thus at the analysis of a level of a number of anti-inflammation in whey of blood of children ВА of a various degree of weight in comparison with a status of remission we mark significant authentic increase of a level of IL-1β  TNFα at heavy attack ВА and moderately increase them at an attack of average weight.  In the easy attack ВА is not revealed of authentic increase of cytokine. The whey level of IL-1β,  TNFα, and IL-4 did not depend on weight current ВА.  In a secret of mouth cavities accepted, that increase of their concentration in comparison by control group. Increase of a level of TNFα in heavy current 2,6 times, average heavy current 1,7 times, easy 1,5 times. The contents IL-4 in heavy current 2,4 times, above control values, average heavy current 1,6 times, easy 1,7 times.  Significant increase of a level of IL-1β in all groups. Thus, a level some anti-inflammation cytokines in biological liquids in the children with bronchial asthma are increased, their contents depend on weight current of disease.
